despite は前置詞で，despite ＋ 名詞（動名詞）の形で「～にもかかわらず」という意味を表します。in spite of ＋ 名詞と同じ意味です。

although との使い分け
despite は前置詞，although は接続詞

× despite of（of がつくのは in spite of）
